Co jsou kanabinoidy?<\/h2>\n<p>Kanabinoidy jsou skupina chemick\u00fdch l\u00e1tek, kter\u00e9 ovliv\u0148uj\u00ed s\u00ed\u0165 receptor\u016f rozprost\u0159enou po cel\u00e9m lidsk\u00e9m t\u011ble i nervov\u00e9m syst\u00e9mu. Nejv\u00edc jsou zn\u00e1m\u00e9 d\u00edky sv\u00e9 p\u0159\u00edtomnosti v rostlin\u011b konop\u00ed, jejich p\u0159\u00edb\u011bh je ale mnohem \u0161ir\u0161\u00ed.<\/p>\n<p>Jen v rostlin\u011b Cannabis sativa v\u011bdci identifikovali p\u0159es 100 r\u016fzn\u00fdch kanabinoid\u016f. Ka\u017ed\u00fd z nich m\u00e1 m\u00edrn\u011b odli\u0161n\u00fd molekul\u00e1rn\u00ed tvar, a pr\u00e1v\u011b ten ur\u010duje, jak bude s receptory v t\u011ble interagovat, jak\u00e9 \u00fa\u010dinky vyvol\u00e1 a jak siln\u00e9 budou.<\/p>\n<p>Kanabinoidy d\u011bl\u00edme do t\u0159\u00ed z\u00e1kladn\u00edch skupin a je dobr\u00e9 je od za\u010d\u00e1tku zn\u00e1t:<\/p>\n<ul>\n<li><strong>Fytokanabinoidy<\/strong> vznikaj\u00ed p\u0159irozen\u011b v rostlin\u00e1ch, p\u0159edev\u0161\u00edm v Cannabis sativa, v men\u0161\u00edm mno\u017estv\u00ed je ale najdeme tak\u00e9 v \u010dern\u00e9m pep\u0159i, t\u0159apatce nebo kakau.<\/li>\n<li><strong>Endokanabinoidy<\/strong> si podle pot\u0159eby vytv\u00e1\u0159\u00ed p\u0159\u00edmo lidsk\u00e9 t\u011blo. Jde o vlastn\u00ed sign\u00e1ln\u00ed molekuly organismu, kter\u00e9 funguj\u00ed ve stejn\u00e9m receptorov\u00e9m syst\u00e9mu.<\/li>\n<li><strong>Syntetick\u00e9 kanabinoidy<\/strong> vznikaj\u00ed v laborato\u0159i, a\u0165 u\u017e pro farmaceutick\u00e9 \u00fa\u010dely, nebo v nebezpe\u010dn\u00e9 podob\u011b jako rekrea\u010dn\u00ed drogy.<\/li>\n<\/ul>\n<p>Kdy\u017e v\u011bt\u0161ina lid\u00ed mluv\u00ed o kanabinoidech, m\u00e1 na mysli pr\u00e1v\u011b fytokanabinoidy, konkr\u00e9tn\u011b ty, kter\u00e9 poch\u00e1zej\u00ed z <a href=\"https:\/\/hempika.com\/cs\/konopi\/\">technick\u00e9ho konop\u00ed<\/a>. Nejzn\u00e1m\u011bj\u0161\u00ed jsou CBD (kanabidiol) a THC (tetrahydrokanabinol). S postupuj\u00edc\u00edm v\u00fdzkumem ale roste v\u011bdeck\u00fd i komer\u010dn\u00ed z\u00e1jem tak\u00e9 o \u0159adu vedlej\u0161\u00edch kanabinoid\u016f, jako jsou CBG, CBN, CBC nebo THCV.<\/p>\n<\/section>\n<section id=\"how-cannabinoids-are-made\">\n<h2>2. Jak kanabinoidy vznikaj\u00ed?<\/h2>\n<p>Kdy\u017e pochop\u00edte, jak rostlina konop\u00ed kanabinoidy vytv\u00e1\u0159\u00ed, sn\u00e1z si vysv\u011btl\u00edte, pro\u010d se jednotliv\u00e9 konopn\u00e9 produkty tolik li\u0161\u00ed vzhledem, v\u016fn\u00ed i \u00fa\u010dinkem, a pro\u010d syrov\u00fd, nezah\u0159\u00e1t\u00fd <a href=\"https:\/\/hempika.com\/cs\/produkt\/konopne-kvety\/\">konopn\u00fd kv\u011bt<\/a> obsahuje jin\u00e9 l\u00e1tky ne\u017e <a href=\"https:\/\/hempika.com\/cs\/produkt\/cbd-kapky\/\">CBD olej<\/a>.<\/p>\n<h3>V\u00fdchoz\u00ed bod: CBGA<\/h3>\n<p>V\u0161echny kanabinoidy vznikaj\u00ed z kanabigerolov\u00e9 kyseliny (CBGA), kter\u00e9 se n\u011bkdy \u0159\u00edk\u00e1 \u201ematka kanabinoid\u016f&#8220;. CBGA vznik\u00e1 chemickou reakc\u00ed dvou jednodu\u0161\u0161\u00edch prekurzor\u016f, olivetolov\u00e9 kyseliny a geranyl-pyrofosf\u00e1tu, uvnit\u0159 trichom\u016f rostliny, tedy drobn\u00fdch \u017el\u00e1znat\u00fdch chloupk\u016f, kter\u00e9 pokr\u00fdvaj\u00ed kv\u011bty a listy.<\/p>\n<p>Z CBGA pak specifick\u00e9 rostlinn\u00e9 enzymy vytv\u00e1\u0159ej\u00ed t\u0159i hlavn\u00ed kysel\u00e9 prekurzory:<\/p>\n<ul>\n<li><strong>CBDA<\/strong> (kanabidiolov\u00e1 kyselina) je prekurzorem CBD<\/li>\n<li><strong>THCA<\/strong> (tetrahydrokanabinolov\u00e1 kyselina) je prekurzorem THC<\/li>\n<li><strong>CBCA<\/strong> (kanabichromenov\u00e1 kyselina) je prekurzorem CBC<\/li>\n<\/ul>\n<p>\u010c\u00e1st CBGA, kterou tyto enzymy nep\u0159em\u011bn\u00ed, se m\u016f\u017ee dekarboxylac\u00ed prom\u011bnit p\u0159\u00edmo v CBG.<\/p>\n<h3>Dekarboxylace: aktivace kanabinoid\u016f<\/h3>\n<p>V syrov\u00e9m, p\u0159irozen\u00e9m stavu existuj\u00ed kanabinoidy jako kyseliny. CBDA nen\u00ed tot\u00e9\u017e co CBD a THCA na rozd\u00edl od THC nem\u00e1 psychoaktivn\u00ed \u00fa\u010dinky. P\u0159em\u011bna z kysel\u00e9 na aktivn\u00ed formu prob\u00edh\u00e1 dekarboxylac\u00ed, tedy od\u0161t\u011bpen\u00edm karboxylov\u00e9 skupiny (COOH), p\u0159i kter\u00e9m se uvol\u0148uje CO<sub>2<\/sub>.<\/p>\n<p>K tomu p\u0159irozen\u011b doch\u00e1z\u00ed, kdy\u017e je rostlinn\u00fd materi\u00e1l konop\u00ed:<\/p>\n<ul>\n<li><strong>Zah\u0159\u00edv\u00e1n<\/strong> (kou\u0159en\u00ed, vapov\u00e1n\u00ed, va\u0159en\u00ed)<\/li>\n<li><strong>Dlouhodob\u011b vystaven sv\u011btlu nebo vzduchu<\/strong> (pomalej\u0161\u00ed proces, kter\u00fd prob\u00edh\u00e1 postupn\u011b)<\/li>\n<\/ul>\n<p>Proto se CBD oleje vyr\u00e1b\u011bn\u00e9 z konopn\u00e9ho extraktu p\u0159i zpracov\u00e1n\u00ed zpravidla dekarboxyluj\u00ed, aby bylo CBD v aktivn\u00ed, vyu\u017eiteln\u00e9 form\u011b. Ze stejn\u00e9ho d\u016fvodu obsahuj\u00ed syrov\u00e9 konopn\u00e9 listy pou\u017e\u00edvan\u00e9 do \u0161\u0165\u00e1v nebo kapsl\u00ed sp\u00ed\u0161 CBDA ne\u017e CBD, a proto se tak\u00e9 n\u011bkter\u00e9 produkty c\u00edlen\u011b vyr\u00e1b\u011bj\u00ed tak, aby si syrov\u00e9 kysel\u00e9 formy zachovaly.<\/p>\n<\/section>\n<section id=\"the-endocannabinoid-system\">\n<h2>3. Endokanabinoidn\u00ed syst\u00e9m<\/h2>\n<p>Endokanabinoidn\u00ed syst\u00e9m (ECS) pat\u0159\u00ed mezi nejroz\u0161\u00ed\u0159en\u011bj\u0161\u00ed a nejd\u016fle\u017eit\u011bj\u0161\u00ed regula\u010dn\u00ed syst\u00e9my lidsk\u00e9ho t\u011bla. Objeven byl ale a\u017e na po\u010d\u00e1tku 90. let, tedy pom\u011brn\u011b ned\u00e1vno, a proto o n\u011bm st\u00e1le v\u00edme m\u00e9n\u011b ne\u017e t\u0159eba o kardiovaskul\u00e1rn\u00edm nebo nervov\u00e9m syst\u00e9mu.<\/p>\n<p>ECS tvo\u0159\u00ed t\u0159i hlavn\u00ed sou\u010d\u00e1sti:<\/p>\n<ol>\n<li><strong>Endokanabinoidy<\/strong>, sign\u00e1ln\u00ed molekuly, kter\u00e9 si t\u011blo vytv\u00e1\u0159\u00ed samo<\/li>\n<li><strong>Kanabinoidn\u00ed receptory<\/strong>, b\u00edlkoviny na povrchu bun\u011bk, na kter\u00e9 se endokanabinoidy i fytokanabinoidy v\u00e1\u017e\u00ed<\/li>\n<li><strong>Enzymy<\/strong>, kter\u00e9 endokanabinoidy podle pot\u0159eby vytv\u00e1\u0159ej\u00ed a po spln\u011bn\u00ed jejich \u00falohy zase odbour\u00e1vaj\u00ed<\/li>\n<\/ol>\n<h3>Receptory CB1<\/h3>\n<p>Receptory CB1 se nach\u00e1zej\u00ed p\u0159edev\u0161\u00edm v mozku a centr\u00e1ln\u00ed nervov\u00e9 soustav\u011b, zejm\u00e9na v oblastech, kter\u00e9 \u0159\u00edd\u00ed pam\u011b\u0165, koordinaci, vn\u00edm\u00e1n\u00ed bolesti a chu\u0165 k j\u00eddlu. Pat\u0159\u00ed mezi v\u016fbec nejpo\u010detn\u011bj\u0161\u00ed receptory v mozku. Kdy\u017e se na n\u011b nav\u00e1\u017ee THC, vyvol\u00e1 to typick\u00e9 psychoaktivn\u00ed \u00fa\u010dinky konop\u00ed. CBD m\u00e1 naproti tomu k CB1 receptor\u016fm jen velmi n\u00edzkou afinitu, a proto omamn\u00e9 \u00fa\u010dinky nezp\u016fsobuje.<\/p>\n<h3>Receptory CB2<\/h3>\n<p>Receptory CB2 se nach\u00e1zej\u00ed hlavn\u011b v imunitn\u00edm syst\u00e9mu a perifern\u00edch tk\u00e1n\u00edch, v\u010detn\u011b st\u0159ev, sleziny, jater a k\u016f\u017ee. Hraj\u00ed kl\u00ed\u010dovou roli p\u0159i regulaci z\u00e1n\u011btu a imunitn\u00ed odpov\u011bdi. L\u00e1tky, kter\u00e9 c\u00edl\u00ed na CB2 receptory, jsou proto p\u0159edm\u011btem zvl\u00e1\u0161tn\u00edho z\u00e1jmu ve v\u00fdzkumu z\u00e1n\u011btliv\u00fdch onemocn\u011bn\u00ed.<\/p>\n<h3>Nejen CB1 a CB2<\/h3>\n<p>V\u00fdzkum uk\u00e1zal, \u017ee kanabinoidy neovliv\u0148uj\u00ed jen tyto dva klasick\u00e9 receptory, ale i \u0159adu dal\u0161\u00edch c\u00edl\u016f:<\/p>\n<ul>\n<li><strong>TRPV1<\/strong> (vaniloidn\u00ed receptor) se pod\u00edl\u00ed na p\u0159enosu bolesti a regulaci t\u011blesn\u00e9 teploty. CBD je zn\u00e1m\u00fdm agonistou TRPV1.<\/li>\n<li><strong>5-HT1A<\/strong> (serotoninov\u00fd receptor) souvis\u00ed s regulac\u00ed n\u00e1lady a \u00fazkosti. Bylo prok\u00e1z\u00e1no, \u017ee ho CBD aktivuje, co\u017e m\u016f\u017ee \u010d\u00e1ste\u010dn\u011b vysv\u011btlovat jeho \u00fa\u010dinky na \u00fazkost.<\/li>\n<li><strong>GPR55<\/strong>, kter\u00e9mu se n\u011bkdy \u0159\u00edk\u00e1 \u201et\u0159et\u00ed kanabinoidn\u00ed receptor&#8220;, ovliv\u0148uje hustotu kost\u00ed, krevn\u00ed tlak a vn\u00edm\u00e1n\u00ed bolesti.<\/li>\n<li><strong>PPAR\u03b3<\/strong> je jadern\u00fd receptor pod\u00edlej\u00edc\u00ed se na metabolismu, z\u00e1n\u011btu a bun\u011b\u010dn\u00e9m d\u011blen\u00ed.<\/li>\n<\/ul>\n<h3>Co ECS vlastn\u011b d\u011bl\u00e1?<\/h3>\n<p>ECS b\u00fdv\u00e1 ozna\u010dov\u00e1n za jak\u00e9hosi hlavn\u00edho regul\u00e1tora, syst\u00e9m, kter\u00fd pom\u00e1h\u00e1 udr\u017eovat t\u011blo v rovnov\u00e1ze (homeost\u00e1ze). Jeho \u00faloha byla zdokumentov\u00e1na v souvislosti s:<\/p>\n<ul>\n<li>Bolest\u00ed a z\u00e1n\u011btem<\/li>\n<li>N\u00e1ladou a emo\u010dn\u00edmi reakcemi<\/li>\n<li>Pam\u011bt\u00ed a u\u010den\u00edm<\/li>\n<li>Chut\u00ed k j\u00eddlu a metabolismem<\/li>\n<li>Sp\u00e1nkov\u00fdm cyklem<\/li>\n<li>Funkc\u00ed imunitn\u00edho syst\u00e9mu<\/li>\n<li>Reakc\u00ed na stres<\/li>\n<li>Neuroprotekc\u00ed<\/li>\n<\/ul>\n<p>D\u016fle\u017eit\u00e1 zvl\u00e1\u0161tnost: na rozd\u00edl od v\u011bt\u0161iny neurotransmiterov\u00fdch syst\u00e9m\u016f funguje ECS obr\u00e1cen\u011b. Kdy\u017e se postsynaptick\u00fd neuron p\u0159ebud\u00ed, vy\u0161le endokanabinoidy zp\u011bt k presynaptick\u00e9mu neuronu, aby sign\u00e1l zpomalil. Pr\u00e1v\u011b tato zp\u011btn\u00e1 (retrogr\u00e1dn\u00ed) signalizace d\u011bl\u00e1 z kanabinoid\u016f jedine\u010dn\u00fd druh biologick\u00e9 brzdy.<\/p>\n<\/section>\n<section id=\"phytocannabinoids\">\n<h2>4. Fytokanabinoidy: hlavn\u00ed hr\u00e1\u010di<\/h2>\n<p>V n\u00e1sleduj\u00edc\u00edm p\u0159ehledu p\u0159edstavujeme nejv\u00fdznamn\u011bj\u0161\u00ed kanabinoidy, kter\u00e9 najdeme v technick\u00e9m konop\u00ed i v konop\u00ed obecn\u011b.<\/p>\n<h3>CBD (kanabidiol)<\/h3>\n<p><a href=\"https:\/\/hempika.com\/cs\/kanabidiol-cbd\/\">CBD<\/a> je dominantn\u00ed kanabinoid leg\u00e1ln\u00edho konop\u00ed a z\u00e1rove\u0148 nejv\u00edc prozkouman\u00e1 neomamn\u00e1 l\u00e1tka, kterou tato rostlina obsahuje. Na klasick\u00e9 CB1 ani CB2 receptory se nev\u00e1\u017ee siln\u011b. ECS ovliv\u0148uje sp\u00ed\u0161 nep\u0159\u00edmo, a to inhibic\u00ed enzymu FAAH (kter\u00fd odbour\u00e1v\u00e1 endokanabinoid anandamid), aktivac\u00ed receptor\u016f TRPV1 a 5-HT1A a jako negativn\u00ed alosterick\u00fd modul\u00e1tor CB1, d\u00edky \u010demu\u017e m\u016f\u017ee p\u0159i sou\u010dasn\u00e9 p\u0159\u00edtomnosti THC jeho \u00fa\u010dinky dokonce tlumit.<\/p>\n<p>V\u00fdzkum CBD je nejpokro\u010dilej\u0161\u00ed ze v\u0161ech kanabinoid\u016f. Nejpevn\u011bj\u0161\u00ed klinick\u00e9 d\u016fkazy existuj\u00ed u epilepsie: farmaceutick\u00fd p\u0159\u00edpravek Epidiolex na b\u00e1zi CBD schv\u00e1lil americk\u00fd \u00fa\u0159ad FDA pro dv\u011b vz\u00e1cn\u00e9 formy d\u011btsk\u00e9 epilepsie, Dravet\u016fv syndrom a Lennox\u016fv-Gastaut\u016fv syndrom. Krom\u011b toho zkoum\u00e1 rozs\u00e1hl\u00e1 \u0159ada preklinick\u00fdch i klinick\u00fdch studi\u00ed \u00fa\u010dinky CBD na \u00fazkost, sp\u00e1nek, z\u00e1n\u011bt, bolest a neuroprotekci.<\/p>\n<p><strong>Pr\u00e1vn\u00ed status:<\/strong> V EU je prodej CBD produkt\u016f vyroben\u00fdch ze schv\u00e1len\u00fdch odr\u016fd konop\u00ed ve form\u011b dopl\u0148k\u016f stravy leg\u00e1ln\u00ed, pokud spl\u0148uj\u00ed na\u0159\u00edzen\u00ed o nov\u00fdch potravin\u00e1ch (novel food) a obsahuj\u00ed m\u00e9n\u011b THC, ne\u017e stanovuje limit dan\u00e9 zem\u011b (obvykle 0,2 a\u017e 0,3 %).<\/p>\n<h3>THC (delta-9-tetrahydrokanabinol)<\/h3>\n<p>THC je hlavn\u00ed psychoaktivn\u00ed kanabinoid konop\u00ed. P\u0159\u00edmo a siln\u011b se v\u00e1\u017ee na CB1 receptory v mozku, \u010d\u00edm\u017e vyvol\u00e1v\u00e1 omamn\u00e9 \u00fa\u010dinky spojovan\u00e9 s marihuanou. V technick\u00e9m konop\u00ed se vyskytuje jen ve stopov\u00e9m mno\u017estv\u00ed. Z\u00e1konn\u00fd limit v EU je 0,3 % v su\u0161en\u00e9m kv\u011btu, hotov\u00e9 v\u00fdrobky pak mus\u00ed obvykle obsahovat m\u00e9n\u011b ne\u017e 0,2 % (podle konkr\u00e9tn\u00ed zem\u011b).<\/p>\n<p>Navzdory sv\u00e9 pov\u011bsti je THC i p\u0159edm\u011btem seri\u00f3zn\u00edho l\u00e9ka\u0159sk\u00e9ho v\u00fdzkumu a je \u00fa\u010dinnou l\u00e1tkou schv\u00e1len\u00fdch farmaceutick\u00fdch p\u0159\u00edpravk\u016f proti nevolnosti (dronabinol) i kombinovan\u00e9 terapie Sativex (v pom\u011bru THC:CBD 1:1) na spasticitu spojenou s roztrou\u0161enou skler\u00f3zou.<\/p>\n<p><strong>Pr\u00e1vn\u00ed status:<\/strong> Ve v\u011bt\u0161in\u011b zem\u00ed kontrolovan\u00e1 l\u00e1tka p\u0159i koncentrac\u00edch nad stanoven\u00fd limit. V leg\u00e1ln\u00edch konopn\u00fdch produktech se vyskytuje jen ve stopov\u00e9m mno\u017estv\u00ed.<\/p>\n<h3>CBG (kanabigerol)<\/h3>\n<p>CBG vznik\u00e1 p\u0159\u00edmou dekarboxylac\u00ed CBGA, \u201ematky kanabinoid\u016f&#8220;, a proto se mu \u010dasto \u0159\u00edk\u00e1 prekurzorov\u00e1 molekula. U vzrostl\u00fdch rostlin konop\u00ed je v\u011bt\u0161ina CBGA u\u017e p\u0159em\u011bn\u011bn\u00e1 na jin\u00e9 kanabinoidy, tak\u017ee se CBG obvykle vyskytuje jen v mal\u00e9m mno\u017estv\u00ed, pokud rostlina nen\u00ed z\u00e1m\u011brn\u011b vy\u0161lecht\u011bn\u00e1 tak, aby si ho zachovala. CBD tento enzym tlum\u00ed, \u010d\u00edm\u017e prodlu\u017euje dobu, po kterou z\u016fst\u00e1v\u00e1 anandamid v organismu aktivn\u00ed.
